Product Description

Naptumomab estafenatox (brand name Naptumomab) is an investigational immunocytokine fusion protein composed of a mouse monoclonal antibody fragment (Fab) targeting the tumor-associated antigen 5T4 (also known as TPBG) linked to a modified superantigen, staphylococcal enterotoxin E (SEE). The 5T4-specific Fab directs the construct to tumor cells expressing 5T4, while the SEE moiety acts as a polyclonal T-cell activator by binding to specific Vβ regions of the T-cell receptor (TCR), causing massive local T-cell proliferation and cytokine release. This redirects pre-existing T cells to the tumor site, inducing an inflammatory anti-tumor response. It has been clinically evaluated for the treatment of advanced renal cell carcinoma (RCC) and non-small cell lung cancer (NSCLC).

5T4 (trophoblast glycoprotein, TPBG) is an oncofetal antigen—a cell-surface glycoprotein that is highly expressed on many carcinomas (e.g., renal, lung, colorectal, ovarian) but has limited expression in normal adult tissues. Its overexpression is associated with poor prognosis, metastasis, and epithelial-mesenchymal transition (EMT).
