Rocatinlimab (AMG 451/KHK4083) is a fully human IgG1 monoclonal antibody that acts as an OX40 antagonist. It targets and blocks OX40 (CD134), a co-stimulatory receptor on activated T cells. By inhibiting OX40-OX40L interaction, it reduces the activation, proliferation, and survival of pathogenic T cells, thereby dampening the immune response. It is specifically developed for the treatment of moderate-to-severe atopic dermatitis, aiming to reduce skin inflammation and pruritus.

Function

OX40 (also known as CD134 or TNFRSF4) is a co-stimulatory receptor belonging to the tumor necrosis factor receptor superfamily (TNFRSF). It is transiently and selectively expressed on activated CD4+ and CD8+ T cells, as well as on regulatory T cells (Tregs) and natural killer (NK) cells. OX40 is not present on naïve or resting T cells.
