APRIL, is a cytokine crucial for B cell survival, differentiation, and class-switching to IgA. In IgAN, APRIL overexpression drives excessive synthesis of Gd-IgA1, which forms immune complexes that deposit in the kidneys, triggering inflammation and fibrosis. By neutralizing APRIL, sibeprenlimab reduces Gd-IgA1 production, potentially decreasing glomerular injury and preserving renal function.
