Product Description

Bemarituzumab (formerly FPA144) is a humanized monoclonal antibody of the IgG1 kappa isotype. It is an anticancer therapeutic designed with a dual mechanism of action. First, it functions as a signaling antagonist by binding to its target, thereby inhibiting the downstream tumor proliferation and survival pathways, such as MAPK and PI3K/AKT. Second, it is Fc-engineered (defucosylated) to enhance its affinity for the Fc gamma RIIIa (CD16a) receptor on Natural Killer (NK) cells, thereby boosting its Antibody-Dependent Cellular Cytotoxicity (ADCC) to directly destroy tumor cells. Bemarituzumab is primarily being investigated for the treatment of FGFR2b positive, unresectable locally advanced or metastatic gastric or gastroesophageal junction (G/GEJ) adenocarcinoma.

Function

Fibroblast Growth Factor Receptor 2b (FGFR2b), a specific splice variant of FGFR2. FGFR2 is a member of the transmembrane receptor tyrosine kinase family. FGFR2b is a cell surface receptor that is highly overexpressed in certain cancers, including approximately 30 percent to 38 percent of gastric tumors. Upon binding to its ligands (such as FGF7 and FGF10), FGFR2b activates intracellular signaling pathways. This abnormal activation is a major oncogenic driver that promotes tumor cell growth, survival, angiogenesis, and metastasis.
