Product Description

Cinrebafusp alfa (PRS-343) is an investigational, high-affinity bispecific antibody developed by Pieris Pharmaceuticals using its proprietary Anticalin® platform technology. It is designed for the treatment of HER2-positive solid tumors. The drug functions by simultaneously targeting two distinct molecules: the tumor-associated antigen HER2 on cancer cells and the co-stimulatory receptor CD137 (4-1BB) on immune cells. This unique design aims to conditionally activate T cells specifically within the tumor microenvironment, thereby enhancing anti-tumor immunity while potentially minimizing systemic toxicity. The use of the Anticalin scaffold also contributes to a reduced risk of immunogenicity.

Function

ERBB2 receptor, more commonly known as Human Epidermal Growth Factor Receptor 2 (HER2). HER2 is a member of the epidermal growth factor receptor (EGFR/ErbB) family of receptor tyrosine kinases. In approximately 15-20% of breast cancers and other solid tumors, the HER2 gene is amplified, leading to protein overexpression on the cancer cell surface. This overexpression drives aggressive tumor growth, proliferation, and survival through constitutive signaling pathways.
