Falbikitug is an investigational, humanized immunoglobulin G1 (IgG1) monoclonal antibody developed for the treatment of solid tumors. It functions as a targeted inhibitor of leukemia inhibitory factor (LIF), a cytokine in the tumor microenvironment. By binding to LIF with high affinity, falbikitug neutralizes its activity, blocking its interaction with the LIF receptor (LIFR) and the co-receptor gp130 on target cells. This blockade inhibits the downstream JAK/STAT signaling pathway, which is associated with multiple tumor-promoting processes. As a result, falbikitug exerts potential immunomodulating and anti-tumor effects, including reversing immunosuppression, inhibiting cancer stem cell self-renewal, and enhancing the efficacy of other cancer therapies.
