Product Description

Lexatumumab (also known as HGS-ETR2) is a fully human monoclonal antibody that specifically targets and activates TRAIL receptor 2 (TRAIL-R2, also known as DR5 or TNFRSF10B), a member of the tumor necrosis factor (TNF) receptor superfamily. As an agonist antibody, it binds to DR5 and mimics the action of the natural ligand TRAIL (TNF-related apoptosis-inducing ligand), thereby inducing apoptosis (programmed cell death) in cancer cells that express the receptor. It was developed as a potential therapeutic agent for the treatment of various advanced solid tumors and hematologic malignancies, based on the premise of selectively triggering cell death in tumors while sparing most normal cells.

Function

Death Receptor 5 (DR5), also known as TRAIL receptor 2 (TRAILR2) or TNFRSF10B. DR5 is a cell surface receptor belonging to the Tumor Necrosis Factor (TNF) receptor superfamily. For efficient signal transduction leading to apoptosis, DR5 molecules must aggregate and undergo higher-order clustering at the cell membrane. This clustering recruits crucial adaptor proteins (like FADD) to form the Death-Inducing Signaling Complex (DISC), which then activates caspases (cysteine-aspartic acid proteases) to execute cell death.
