Linavonkibart (also known as SRK-181) is a fully human monoclonal antibody designed to selectively bind to and inhibit the activation of latent transforming growth factor-beta 1 (TGFβ1). Unlike antibodies that target the active form of TGFβ or its receptors, linavonkibart specifically interacts with the latent complex, preventing its proteolytic activation and subsequent release of mature TGFβ1. This selective inhibition aims to block the TGFβ1-mediated immunosuppressive pathways within the tumor microenvironment while potentially minimizing systemic toxicity. It is under clinical investigation as a cancer immunotherapy, particularly in combination with immune checkpoint inhibitors (e.g., anti-PD-1), for the treatment of advanced solid tumors.
