Nolavetbart is a caninized IgG2-kappa monoclonal antibody specifically developed for veterinary use. It targets the canine interleukin-31 receptor alpha (IL-31RA), a key component of the receptor for the pruritic (itch-inducing) cytokine IL-31. As an IgG2 isotype, it is engineered to minimize effector functions (e.g., reduced risk of antibody-dependent cellular cytotoxicity) while focusing on receptor blockade. By binding to canine IL-31RA, nolavetbart prevents IL-31 from activating its receptor, thereby inhibiting the downstream signaling pathway that triggers pruritus and inflammation.

Function

Interleukin-31 receptor alpha (IL-31RA) is the specific alpha subunit of the heterodimeric IL-31 receptor complex, which also includes the oncostatin M receptor beta (OSMRβ). This receptor is expressed on sensory neurons, keratinocytes, and immune cells in dogs. Binding of IL-31 (produced mainly by activated Th2 cells) to IL-31RA activates JAK/STAT, MAPK, and PI3K pathways, leading to the sensation of pruritus (itch), skin barrier dysfunction, and inflammation.
