Product Description

Orilanolimab (also known as SYNT001 or ALXN1830) is a humanized IgG4-kappa monoclonal antibody that functions as a neonatal Fc receptor (FcRn) antagonist. By binding with high affinity to FcRn, it competitively blocks the interaction between FcRn and the Fc region of immunoglobulin G (IgG). This inhibition disrupts the normal recycling of IgG, leading to accelerated degradation of endogenous IgG (including pathogenic autoantibodies and alloantibodies) and a reduction in circulating IgG levels.

Function

The neonatal Fc receptor (FcRn) is a MHC class I-like receptor expressed on endothelial cells, phagocytes, and epithelial cells. Its primary physiological role is to protect IgG and albumin from lysosomal degradation, thereby extending their serum half-life (∼3 weeks for IgG). FcRn binds to the Fc region of IgG in a pH-dependent manner (binding in acidic endosomes, release at neutral pH).
