Tisotumab vedotin is an antibody-drug conjugate (ADC) targeting tissue factor (TF, also known as CD142), a transmembrane protein overexpressed in many solid tumors. It consists of a human monoclonal antibody linked to the cytotoxic payload monomethyl auristatin E (MMAE). Upon binding to TF, the ADC is internalized, releasing MMAE to disrupt microtubule assembly and induce apoptosis. It is approved for the treatment of recurrent or metastatic cervical cancer, and is being studied in other TF-positive cancers.
