Angiopoietin-2 is a cytokine that primarily signals through the Tie2 receptor on endothelial cells. Under pathological conditions such as in wet AMD or diabetic retinopathy, elevated Ang2 levels antagonize the stabilizing effects of Angiopoietin-1, promoting vascular destabilization, inflammation, and pathological angiogenesis (sprouting of abnormal, leaky blood vessels). By neutralizing Ang2, zansecimab aims to restore vascular stability and suppress abnormal vessel growth in the eye.
