Product Description

Batoclimab is a fully human monoclonal antibody designed as a high-affinity antagonist of the neonatal Fc receptor (FcRn). It functions by competitively binding to FcRn at the acidic pH of the endosome, thereby blocking the recycling of IgG antibodies (including pathogenic autoantibodies) and accelerating their lysosomal degradation. This leads to a rapid and sustained reduction in circulating levels of all IgG subtypes. It is in clinical development for the treatment of various IgG-driven autoimmune diseases, including myasthenia gravis, thyroid eye disease, chronic inflammatory demyelinating polyneuropathy (CIDP), and warm autoimmune hemolytic anemia (wAIHA).

Function

neonatal Fc receptor (FcRn), a major histocompatibility complex (MHC) class I-like receptor expressed widely on endothelial cells, myeloid cells, and epithelial cells. FcRn plays a crucial role in regulating IgG homeostasis. It binds to IgG in acidic endosomes (pH ~6.0) after pinocytosis, diverts it from lysosomal degradation, and recycles it back to the cell surface where it is released into the circulation at neutral pH. This recycling mechanism is responsible for the exceptionally long half-life of IgG antibodies (approx. 21 days). In autoimmune diseases, this same process inadvertently extends the half-life of pathogenic IgG autoantibodies.
