Golimumab is a fully human monoclonal IgG1κ antibody approved for the treatment of several chronic immune-mediated inflammatory diseases. It functions as a tumor necrosis factor-alpha (TNF-α) inhibitor. By binding with high affinity to both soluble and transmembrane forms of TNF-α, golimumab neutralizes its biological activity, preventing TNF-α from interacting with its receptors, TNFR1 and TNFR2, on immune and tissue cells. This blockade inhibits the downstream pro-inflammatory signaling cascade that drives chronic inflammation, tissue damage, and symptoms in conditions like rheumatoid arthritis, psoriatic arthritis, ankylosing spondylitis, ulcerative colitis, and plaque psoriasis.
