Luvagrobart is a humanized IgG1-kappa (IgG1κ) monoclonal antibody that specifically targets nerve growth factor (NGF), a key neurotrophin involved in pain signaling. As an IgG1 isotype, it is designed to potentially engage effector functions. By binding to and neutralizing NGF, it blocks the interaction between NGF and its high-affinity receptor TrkA (tropomyosin receptor kinase A) on nociceptive neurons. This inhibition aims to reduce the sensitization and hyperexcitability of pain-sensing neurons, thereby alleviating chronic pain. It is being developed as a non-opioid analgesic for the treatment of moderate-to-severe chronic pain conditions, such as osteoarthritis pain and chronic low back pain.
