Nelistotug (also known as GSK6097608) is a human IgG1-kappa monoclonal antibody that specifically targets CD96 (T-cell-activated increased late expression, TACTILE), an immune checkpoint receptor belonging to the immunoglobulin superfamily. As an IgG1 isotype, it is designed to engage immune effector functions such as antibody-dependent cellular cytotoxicity (ADCC) and/or block receptor-ligand interactions. CD96 is primarily expressed on natural killer (NK) cells and T cells, where it binds to ligands such as CD155 (PVR) and CD111 (Nectin-1), often transmitting inhibitory signals. By binding to CD96, nelistotug aims to block these interactions, thereby reversing immune suppression and enhancing NK-cell and T-cell anti-tumor activity. It is being developed as an immune checkpoint inhibitor for the treatment of advanced solid tumors.
