Product Description

Onvatilimab (also known as JNJ-61610588 or CI-8993) is a humanized IgG1-kappa monoclonal antibody that targets V-domain Ig suppressor of T-cell activation (VISTA), an inhibitory immune checkpoint protein. As an IgG1 isotype, it is designed to potentially engage immune effector functions such as antibody-dependent cellular cytotoxicity (ADCC). By binding to VISTA, onvatilimab blocks its interaction with ligands (e.g., VSIG3, PSGL-1), thereby reversing VISTA-mediated suppression of T-cell activation and cytokine production. It is being developed as an immune checkpoint inhibitor for the treatment of advanced solid tumors, with the aim of restoring anti-tumor immunity, particularly in combination with other immunotherapies.

Function

V-domain Ig suppressor of T-cell activation (VISTA), also known as PD-1H or B7-H5, is a type I transmembrane protein belonging to the B7/CD28 family of immune checkpoints. It is predominantly expressed on myeloid cells (e.g., macrophages, dendritic cells, neutrophils) and a subset of T cells. VISTA functions as both a ligand that suppresses T-cell receptor signaling and a receptor that transmits inhibitory signals.
