Tomaralimab (OPN-305) is a humanized IgG4 monoclonal antibody targeting Toll-like receptor 2 (TLR2), a pattern recognition receptor involved in innate immune activation. It binds to TLR2 to block its interaction with pathogen- and damage-associated molecular patterns (PAMPs/DAMPs), thereby inhibiting downstream pro-inflammatory signaling. It is being developed for the treatment of inflammatory and autoimmune diseases, such as rheumatoid arthritis, atherosclerosis, and organ transplant ischemia-reperfusion injury.
