Product Description

Fremanezumab is a fully humanized monoclonal IgG2a antibody approved for the preventive treatment of migraine in adults. It belongs to the class of calcitonin gene-related peptide (CGRP) pathway inhibitors. Fremanezumab functions as a highly selective ligand inhibitor, specifically binding to the calcitonin gene-related peptide (CGRP) ligand itself. By forming complexes with both the α- and β-isoforms of CGRP in the circulation, it prevents CGRP from binding to its receptor. This blockade inhibits the neuroinflammatory cascade and vasodilation believed to underlie migraine attacks, thereby reducing the frequency and severity of migraines.

Function

Calcitonin gene-related peptide (CGRP), specifically the alpha (α) isoform of CGRP. CGRP is a potent neuropeptide that is widely expressed in the peripheral and central nervous systems. It plays a crucial role in the pathophysiology of migraine. During a migraine attack, CGRP is released from activated trigeminal sensory nerves. It then causes vasodilation of cranial blood vessels, transmits pain signals, and promotes neurogenic inflammation in the meninges. CGRP binds to a complex receptor consisting of the calcitonin receptor-like receptor (CLR) and receptor activity-modifying protein 1 (RAMP1).
