The glucagon receptor is a G-protein-coupled receptor primarily expressed in the liver. Its activation by the hormone glucagon stimulates hepatic glucose production (glycogenolysis and gluconeogenesis), playing a critical counter-regulatory role against insulin. In T1D, relative glucagon excess contributes to hyperglycemia and glycemic instability. By antagonizing GCGR, volagidemab aims to suppress excessive hepatic glucose output, thereby lowering blood glucose levels and potentially widening the therapeutic window for insulin therapy.
